New file server: advice on eliminating file locking

Forgive me if I am asking too obvious of a question, but for your Cent machines, are you using the "nolock" option in your mount? What is the OS of the file server? Or is it a storage system like a NetApp or Isilon cluster? Are there options on the server that can be tweaked?
